SYNTHESIS OF SUBSTITUTED ANALOGS OF LAVENDAMYCIN AND OF CONFORMATIONALLY RESTRICTED ANALOGS

Résumé

Lavendamycin is a natural product isolated in 1981 from fermentation broths of Streptomyces lavendulae. This pentacyclic molecule displays antimicrobial, cytotoxic and anticancer properties. Indeed, it inhibits enzymatic activity of topoisomerases I (enzymes playing role in DNA replication). However, its interest is limited by its toxicity for human organism. Previous work carried out in our laboratory led to synthesis of lavendamycin analogs. Unfortunately, although less toxic, these diversely substituted analogs showed a lower biological activity. In these compounds, the free rotation around the bond between B and C rings brings about a conformational mobility. Therefore, we thought interesting to confer a structural rigidity to these molecules by creating a new F ring, by analogy with the structure of camptothecin (the most known inhibitor substance of topoisomerase I).

In order to synthesize conformationally restricted analogs of lavendamycin, we prepared a new pentacyclic analog bearing a hydroxyl group in 3 position. The key-step of this synthesis is a Pictet-Spengler reaction between a functionalised quinoleine and tryptophane methyl ester. On the one hand, we tried to create a new ring via an intramolecular cyclization involving the pyrrolic nitrogen of D ring and various nucleofuge groups, in basic medium. Unfortunately, the preparation of the desired compounds bearing this nucleofuge group proved to be very difficult and this approach did not succeed. On the other hand, an intramolecular cyclization performed between an aldehyde (obtained from hydroxymethyl group) and the secondary amino group of D ring allowed to form the expected hexacyclic structure. Two first conformationally restricted analogs of lavendamycin have thus been obtained in eight or nine steps in 9,9% or 5,1% overall yield.

The conformationally restricted analogs thus obtained were then functionalized. Analog bearing an amino substituent group on A ring in 7 position was firstly obtained. This nitrogen functionality had to be introduced early in the synthesis and a protection into ditosylamide proved to be necessary. We also synthesized analogs with a methoxy group on A ring at 6 position or an ethyl group on B ring at 4 position together with the amino group in 7 position. After circumventing several problems, we achieved to prepare the three analogs series in nine or ten steps in overall yield ranging from 1,2% to 4,8%.

These conformationally restricted analogs of lavendamycin, and some of their precursors, have been subjected to biological tests by the NCI (National Cancer Institute). Several of them showed a moderate biological activity.
La lavendamycine est une molécule d'origine naturelle isolée en 1981 et formée par Streptomyces lavendulae qui est une bactérie. Cette molécule pentacyclique possède des propriétés antivirale, antimicrobienne et anticancéreuse. En effet, elle permet d'inhiber l'activité enzymatique des topoisomérases de type I (enzymes nécessaires à la réplication de l'ADN lors de la division cellulaire). Cependant, son intérêt est assez limité du fait de sa toxicité élevée pour l'organisme humain. Des travaux antérieurs réalisés au laboratoire ont permis de synthétiser des analogues de la lavendamycine. Ces analogues diversement substitués se sont révélés moins toxiques mais de faible activité biologique. Ces composés possèdent une certaine mobilité conformationnelle (rotation autour de liaison entre les cycles). Il serait alors intéressant de conférer une rigidité structurale à ces molécules par la création d'un nouveau cycle afin d'obtenir des structures analogues à celle de la camptothécine (substance inhibitrice la plus connue des topoisomérases de type I).

Pour synthétiser ces analogues de la lavendamycine à géométrie contrainte, nous avons d'abord préparé un premier analogue pentacyclique possédant un groupement hydroxyméthyle en position 3. L'étape-clef de cette synthèse est une réaction de Pictet-Spengler entre une quinoléine fonctionnalisée et l'ester méthylique du tryptophane. Dans un premier temps, nous avons tenté de créer le nouveau cycle via une cyclisation intramoléculaire en milieu basique au niveau d'un nucléofuge (introduit à partir du groupe hydroxyle) et de l'azote pyrrolique du cycle D. Mais la préparation des composés à groupements nucléofuges a posé de gros problèmes et cette approche n'a pas abouti. En revanche dans un deuxième temps, une cyclisation intramoléculaire réalisée à partir d'un aldéhyde (obtenu à partir du groupement hydroxyméthyle) et de l'azote pyrrolique du cycle D a permis de former la structure hexacyclique souhaitée. Les deux premiers analogues de la lavendamycine à géométrie contrainte ont ainsi été obtenus en huit et neuf étapes avec des rendements globaux de 9,9% et 5,1%.

Nous avons poursuivi notre étude par une fonctionnalisation des analogues à mobilité conformationnelle réduite. En effet, nous avons d'abord préparé des analogues possédant un groupement amino en position 7 sur le cycle A. Cette fonctionnalité a été introduite très tôt dans la synthèse et elle a nécessité une protection sous forme de ditosylamide. Nous avons également préparé des analogues qui tout en conservant ce groupement amino en position 7 avaient soit un groupement méthoxy en position 6 sur le cycle A, soit un groupement éthyle sur la position 4 du cycle B. Malgré des difficultés, ces trois séries d'analogues ont été préparées en neuf ou dix étapes avec des rendements globaux compris entre 1,2% et 4,8%.

Ces analogues de la lavendamycine à géométrie contrainte, ou non, ont été soumis à des tests biologiques par le NCI (National Cancer Institute). Ils ont montré généralement une certaine activité biologique mais qui n'était pas suffisamment importante pour être exploitée en tant qu'agents anticancéreux.
