Towards a better understanding of the role of ITM2B in the retina

Résumé

Our group identified a missense mutation in ITM2B underlying a novel autosomal dominant retinal dystrophy (RD) with inner retinal dysfunction and progressive retinal degeneration. The work presented here aim to better understand the function of ITM2B in the retina, in normal and mutated conditions. Through clinical follow up, we demonstrated the slow progression of the disease which is better characterized with functional test. Proteomic experiments performed on human retinal protein extracts showed a unique ITM2B retinal interactome suggesting that ITM2B may have a specific role in this tissue. In addition, our data suggested a potential role of ITM2B in the mitochondria. Interestingly, this finding correlates with some clinical aspects of the disease. Finally, we used induced pluripotent stem cells derived retinal organoids to perform disease modeling and approach the potential pathological mechanisms involved in RD. Our results showed a specific localization of BRI23 (ITM2B secreted fragment), inside the rosettes of the organoids where the photoreceptor mature. Furthermore, we demonstrated a significantly increased expression of rhodopin and a decreased expression of opsin in RD organoids. In conclusion, this complex disorder may be the combination of different factors. Indeed, two different hypotheses co exists: mitochondrial dysfunction, and the toxic accumulation of BRI23 mutant. However, both hypotheses are not exclusive and further investigations will be needed to better understand this peculiar retinal dystrophy.
Notre groupe a identifié par séquençage haut débit, une mutation sur le gène ITM2B dans une famille atteinte d’une nouvelle dystrophie rétinienne autosomique dominante au phénotype unique. L’objectif de ce travail de thèse consiste à mieux comprendre le rôle d’ITM2B dans la rétine. Dans un premier temps, nous avons démontré que l’évolution de la pathologie est lente et est davantage observable via des tests fonctionnels. Puis, des expériences de protéomique effectuées sur de la rétine humaine ont pu mettre en évidence un rôle spécifique d’ITM2B dans la rétine. De plus, nos données suggèrent aussi un rôle d’ITM2B dans la mitochondrie. Ce résultat est d’autant plus intéressant qu’il corrèle avec certains aspects de la clinique. Enfin, nous avons aussi réalisé un modèle de la pathologie à partir de cellules souches pluripotentes de patients et d’apparentés non atteints. Les organoïdes rétinien ainsi générés, ont montrés une accumulation de BRI23 (fragment sécrété d’ITM2B) au niveau des rosettes, où les photorécepteurs vont se développer et devenir mature. Ces expériences nous ont aussi permis de montrer une augmentation de l’expression de rhodopsin et une diminution de l’expression d’opsin dans les organoïdes mutés. En conclusion, deux hypothèses différentes co-existent : dysfonction mitochondriale et possible accumulation toxique du mutant BRI23. Toutefois ces deux hypothèses ne sont pas exclusives. Des expériences supplémentaires seront néanmoins nécessaires pour mieux comprendre cette dystrophie rétinienne atypique.
