Résumé

The JAK-STAT signaling pathway plays a central role in immunology and development through its involvement in major cellular processes (proliferation, differentiation, migration, apoptosis). Although knowledge of its function, targets and regulation is growing, many mechanisms remain incomplete. The JAK-STAT pathway is conserved in many metazoans, both at the functional and molecular levels, and Drosophila is a model of choice for the study of this signaling pathway because the different types of actors composing the pathway show little redundancy. In particular, during oogenesis, the JAK-STAT pathway is involved in various morphogenesis events necessary for egg maturation, offering as many phenotypes to study in order to understand its functioning. In this context, my thesis project aimed to participate in the study of the regulation of the JAK-STAT pathway using the Drosophila ovary as a system. In the egg chamber, or follicle, polar cells, located in pairs at the anterior and posterior poles of the follicular epithelium, are the only JAK-STAT pathway ligand producing cells. Therefore, following ligand secretion, the JAK-STAT signaling was previously described as symmetric between the anterior and posterior poles. By precisely characterizing the JAK-STAT pathway activation pattern in wild type condition, I observed and described that while symmetry does indeed apply at the beginning of oogenesis, an asymmetry is established between the poles by a decrease in the size of the gradient at the posterior pole at mid oogenesis. Fortuitously, I observed that E-Cadherin, a cell-cell adhesion protein, shows similar dynamics in posterior follicle cells. I was able to demonstrate that E-Cadherin is involved in the same cellular processes as the JAK-STAT pathway during oogenesis and that its expression is necessary for transduction of the JAK-STAT pathway. Further study revealed that E-Cadherin forms a complex with one of the actors of the JAK-STAT pathway, the transcription factor Stat92E. Together, these results demonstrated that E-cadherin acts as a positive regulator of the JAK-STAT pathway in the follicular epithelium. In parallel, I investigated the causes and physiological roles of the restriction of the JAK-STAT pathway in posterior follicle cells. The posterior pole of the follicle is characterized by the presence of the oocyte, secreting the Gurken ligand of the EGFR pathway and the combined activation of the EGFR and JAK-STAT pathways in posterior follicle cells induces their differentiation. These cells send back an information of unknown nature to the oocyte, allowing the initiation of the polarized transport and anchoring of the determinant oskar to the posterior cortex of the oocyte. I identified that the decrease in E-Cadherin in posterior follicle cells is mediated by the Gurken/EGFR pathway. I found that the resulting restriction of the JAK-STAT pathway activity gradient allows the differentiation of a subpopulation of posterior follicle cells, which I named Posterior Anchoring Cells (PACs). I observed that the PACs are the only cells that maintain close contact with the posterior membrane of the oocyte between stages 9 and 10b, and that this contact is mainly mediated by E-Cadherin, a target of the JAK-STAT pathway in the PACs. Finally, I also described that, due to this close contact with the oocyte, the number of PACs must be finely regulated as it directly controls the size of the zone to which oskar will be transported and anchored. This new level of regulation of the oskar anchoring zone is crucial, as I have identified that a too wide oskar anchoring zone in the oocyte leads to defects during the development of the future embryo.
La voie de signalisation JAK-STAT joue un rôle central en immunologie et lors du développement par son implication dans des processus cellulaires majeurs (prolifération, différenciation, migration, apoptose). Bien que les connaissances sur son fonctionnement, ses cibles et sa régulation s'étoffent, nombreux sont les mécanismes restants incomplets. Elle est conservée chez de nombreux métazoaires, autant au niveau fonctionnel que moléculaire et la drosophile est un modèle de choix pour l'étude de cette voie de signalisation car les différents types d'acteur la composant présentent peu de redondance. En particulier, au cours de l'ovogenèse, la voie JAK-STAT est impliquée dans de nombreux évènements de morphogenèse nécessaires à la maturation de l'œuf, offrant autant de phénotypes à étudier pour comprendre son fonctionnement. Dans ce contexte, mon projet de thèse avait pour but de participer à l'étude de la régulation de la voie JAK-STAT en utilisant comme système l'ovaire de drosophile. Au niveau de la chambre à œuf, ou follicule, les cellules polaires, localisées en paires aux pôles antérieur et postérieur de l'épithélium folliculaire, sont les seules productrices du ligand de la voie JAK-STAT. De fait, suite à la sécrétion du ligand, la signalisation était jusqu'ici décrite comme symétrique entre les pôles antérieur et postérieur. En caractérisant précisément le patron d'activité de la voie JAK-STAT en contexte sauvage, j'ai observé et décrit que si la symétrie s'applique en effet au début de l'ovogenèse, une asymétrie se met en place entre les pôles par une diminution de la taille du gradient au pôle postérieur en milieu d'ovogenèse. De façon fortuite, j'ai observé que la E-Cadhérine, une protéine d'adhérence inter-cellulaire, présente une dynamique similaire dans les cellules folliculaires postérieures. J'ai pu démontrer que la E-Cadhérine est impliquée dans les mêmes processus cellulaires que la voie JAK-STAT lors de l'ovogenèse et que son expression est nécessaire à la transduction de la voie JAK-STAT. La poursuite de cette étude a mis en évidence que la E-Cadhérine forme un complexe avec un des acteurs de la voie JAK-STAT, le facteur de transcription Stat92E. L'ensemble de ces résultats a permis de démontrer que la E-cadhérine agit comme régulateur positif de la voie JAK-STAT dans l'épithélium folliculaire. En parallèle, j'ai étudié les causes et rôles physiologiques de la restriction de la voie JAK-STAT dans les cellules folliculaires postérieures. Le pôle postérieur du follicule est caractérisé par la présence de l'ovocyte, sécrétant le ligand Gurken de la voie EGFR et l'activation combinée des voies EGFR et JAK-STAT dans les cellules folliculaires postérieures induit leur différenciation. Ces cellules envoient en retour une information de nature inconnue vers l'ovocyte, permettant l'initiation du transport polarisé et de l'ancrage du déterminant oskar au cortex postérieur de l'ovocyte. J'ai identifié que la diminution de la E-Cadhérine dans les cellules folliculaires postérieures est exercée par la voie Gurken/EGFR. J'ai constaté que la restriction résultante du gradient d'activité de la voie JAK-STAT permet la différenciation d'une sous-population de cellules folliculaires postérieures, que j'ai nommée cellules d'ancrage postérieur. J'ai observé que ces cellules sont les seules gardant un contact étroit avec la membrane postérieure de l'ovocyte entre les stades 9 et 10b et ce grâce à la E-Cadhérine, qui est une cible de la voie JAK-STAT dans les cellules d'ancrage postérieur. Enfin, j'ai également décrit que, du fait de ce contact avec l'ovocyte, le nombre de cellules d'ancrage postérieur doit être finement régulé car il contrôle directement la taille de la zone vers laquelle va être transporté et s'ancrer oskar. Ce nouveau niveau de régulation de la zone d'ancrage d'oskar est crucial, car j'ai identifié qu'une zone d'ancrage trop grande dans l'ovocyte mène à des défauts lors du développement du futur embryon.
