Phaseless diffraction tomography in the multiple scattering regime

Optical diffraction tomography is an imaging technique in which a target is
successively illuminated by a laser beam under different angles of incidence with
an interferometric setup which enables to record both the phase and the intensity
of the field in the image space of a microscope. The permittivity of the target is
then numerically estimated from the complex field dataset thanks to an inversion
procedure.
In this typescript, we first study the effect of multiple scattering inside the target
on the obtained reconstructions by studying different approximations (Born) to speed
up the computation.
The recording of the phase in the optical domain is tricky, for it requires an
interferometric setup. I have built a reconstruction technique based only on the
intensities. I show that the obtained reconstructions are close to those obtained with
the complete dataset field + phase.
Finally, we propose to adjoin phaseless diffraction tomography to fluorescence
structured illumination microscopy. In this framework, we illustrate the impact of
the modelization of the illumination excitating the fluorophores on the resolving
power of the imaging technique. Moreover, we demonstrate an imaging method which
estimates simultaneously the fluorophores and the surrounding media from the joint
recording of diffraction and fluorescence intensities.
La tomographie de diffraction optique est une technique d’imagerie dans laquelle
un objet est illuminé successivement par un faisceau laser sous différents angles
d’incidence avec une méthode interférométrique qui permet d’enregistrer à la fois la
phase et l’intensité du champ dans l’espace image d’un microscope. La permittivité de
l’objet est alors reconstruite numériquement à partir du jeu de données des champs
complexes à l’aide d’une méthode d’inversion.
Dans ce tapuscript, nous étudions tout d’abord l’effet de la diffusion multiple
dans l’objet sur les reconstructions obtenues, en étudiant notamment différentes
possibilités d’approximation (Born) pour accélérer le calcul.
La mesure de la phase dans le domaine de l’optique étant délicate, car obtenue par
une méthode interférentielle, j’ai développé une technique de reconstruction basée
uniquement sur les intensités. J’ai notamment montré que les reconstructions ainsi
obtenues sont proches de celles avec le jeu de données complet champ + phase.
Enfin, nous proposons de coupler la tomographie de diffraction sans mesure de
phase à la microscopie de fluorescence à illumination structurée. Dans ce cadre, nous
illustrons l’impact de la modélisation de l’illumination excitatrice au niveau des
fluorophores sur le pouvoir de résolution de la technique d’imagerie. Par ailleurs,
nous présentons une méthode d’imagerie qui estime simultanément les fluorophores
et le milieu qui les entoure à partir de la mesure conjointe des intensités de diffraction
et de fluorescence.
