Résumé

Owing to the unique properties of the trifluoromethyl (CF₃) group, CF₃-substituted alkenes play important roles in pharmaceuticals, agrochemicals, and materials science. Among various trifluoromethyl alkene derivatives, CF₃-containing acrylates and acrylic acids have great application potential. Despite obvious interest in such kinds of scaffolds, the methods for their synthesis as well as their substrate scope remain relatively limited. In this Ph.D. thesis, we mainly focused on the synthesis of 3-aryl-2-(trifluoromethyl)acrylates and acrylic acids. Subsequent transformations of these compounds were further investigated. In the first part of this thesis, we reported the synthesis of diverse methyl and tert-butyl 3-aryl-2-(trifluoromethyl)acrylates from aryl iodides and the corresponding 2-(trifluoromethyl)acrylates by Mizoroki-Heck reaction in the presence of palladium catalyst and silver(I) additive. The one-pot synthesis of 3-trifluoromethyl coumarin derivatives was further achieved starting from ortho-iodophenols and methyl 2-(trifluoromethyl)acrylate by tandem Mizoroki-Heck reaction and intramolecular cyclization. In the second part of this thesis, we demonstrated that the palladium-catalysed silver(I) additive-mediated Mizoroki-Heck reaction could be extended to the synthesis of 3-aryl-2-(trifluoromethyl)acrylic acids. Similarly, the reaction between ortho-iodophenols and 2-(trifluoromethyl)acrylic acid could also give access to 3-trifluoromethyl coumarin derivatives. Furthermore, we achieved the construction of more complex alkyl 3-aryl-2-(trifluoromethyl)acrylates by three-component reactions between aryl iodides, 2-(trifluoromethyl)acrylic acid, and alkyl iodides. In the last part of this thesis, we developed a method for palladium-catalysed copper(I)-mediated oxidative decarboxylative cross-coupling of heteroarenes with (Z)-3-aryl-2-(trifluoromethyl)acrylic acids. Diverse trisubstituted CF₃-alkenes were generated in relatively low yields but with high E/Z selectivity.
En raison des propriétés uniques du groupement trifluorométhyle (CF₃), les alcènes substitués par un CF₃ jouent un rôle important en chimie pharmaceutique, agrochimie et science des matériaux. Parmi les divers dérivés trifluorométhylés, les acrylates et les acides acryliques contenant un groupement CF₃ présentent un grand potentiel d’applications. Malgré l’intérêt évident de tels « briques de construction », leur méthode de synthèse ainsi que leur étendue d’applications restent relativement limitées. Au cours de cette thèse, nous nous sommes concentrés sur la synthèse des 3-aryl-2-trifluorométhylacrylates ainsi que des acides acryliques correspondants. Des transformations ultérieures de ces composés ont également été étudiées. Dans la première partie de la thèse, nous présentons les résultats obtenus pour la synthèse de divers 3-aryl-2-trifluorométhylacrylates de méthyle ou de tert-butyle. Ces produits trifluorométhylés ont été obtenus à partir de composés aromatiques iodés et des 2-trifluorométhylacrylates correspondants par une réaction de Mizoroki-Heck en présence d’un catalyseur au palladium et d’un additif à base d’argent (I). La synthèse « en un seul pot » de 3-trifluorométhyl coumarines a également été effectuée à partir des ortho-iodophénols et du 2-trifluorométhylacrylate de méthyle par une réaction tandem de Mizoroki-Heck et de cyclisation intramoléculaire. Dans la seconde partie de la thèse, nous avons démontré que la réaction de Mizoroki-Heck catalysée au palladium en présence d’additif à base d’argent pouvait être étendue à la synthèse des acides acryliques 3-aryl-2-trifluorométhylés. De façon similaire, la réaction entre des ortho-iodophénols et de l’acide acrylique 2-trifluorométhylé donne accès aux coumarines 3-trifluorométhylés. De plus, nous avons effectué la construction d’acrylates 3-aryl-2-trifluorométhylés d’alkyle plus complexes par des réactions multi-composants entre des iodures aromatiques, de l’acide 2-trifluorométhyl acrylique et des iodoalcanes. Dans la dernière partie de la thèse, nous avons développé une méthode de couplage croisé décarboxylatif oxydatif catalysé par le palladium en présence de sel de cuivre (I) entre des hétéroarènes et des acides acryliques 3-aryl-2-trifluorométhylés. Divers alcènes trisubstitués ont été obtenus en rendements relativement faibles mais avec une sélectivité E/Z élevée.
