CNTFET-based biosensor technology for kinase activity monitoring

Développement d'un biocapteur à base de transistors en nanotubes de carbone

Résumé

The thesis objective is the conception of a new technology of biosensors based on carbon nanotubes. This conception aims to the integration of these nanostructures as transducers in a chemical gate and as a base for a field effect transistor architecture. The chemical gate and transistor architecture will be designed with the goal of field effect transduction. The target chosen for this technology is the kinase enzyme family. More specifically, the thesis aims to the design and realization of a proof of concept for a specific target: the complex CDK4/cyclinD.The research is threefold. First, the work consists in the design of the new architecture based on literature. Then the second objective is to design a clean room process for manufacturing of the architecture. Finally, the goal of the thesis is to realize manufacturing of the sensor to the proof of concept.This new technology is based on an existing chemical sensor that demonstrated selectivity and sensitivity. The current design aims to reduce human interaction and the use of heavy equipment during measurements. The second design constraint is the passivity of the sensor toward the biological field containing its target. This would allow monitoring of the target and integration in a multisensor architecture.The clean room process aims to be robust and reproducible for future integration of the sensor in an acquisition chain able to generate large quantities of normalized data. The second goal of the process design is to allow the production of large series of sensors at low unitary cost.This thesis manuscript presents the research work leading to the design of the new architecture followed by the design of the manufacturing process. Then the realization results are presented with the trials leading to the characterization of CNTFETs readily modified for selective biosensing.
L'objectif de la thèse est la conception d'une nouvelle technologie de biocapteurs basée sur l'utilisation de nanotubes de carbones. Cette conception vise à utiliser ces nanostructures en tant qu'éléments transducteurs grâce à la conception d'une grille chimique et amplificateurs grâce à leur intégration dans une architecture de transistor à effet de champ. La grille chimique de la structure transistor sera conçue pour être sélective et en concordance avec la transduction de l’effet de champ. Dans ce but, la cible choisie est la famille des enzymes kinases. Le travail de thèse focalise sur la conception et réalisation d’une preuve de concept pour la technologie complète en visant une enzyme kinase spécifique.Le travail de recherche se découpe en trois temps. Tout d'abord, le travail consiste en la définition de l'architecture du capteur. Ensuite, partant de cette définition, il est nécessaire de concevoir un procédé de fabrication dédié. Enfin, le capteur peut être réalisé, si possible jusqu'à la démonstration d'une preuve de concept.La nouvelle technologie se base sur un biocapteur existant dont la sélectivité a été démontrée. La conception du capteur vise à alléger la mesure en temps humain et éviter l'utilisation d'équipements lourds. Nous sommes imposés comme contrainte que le capteur soit passif par rapport à l'échantillon biologique. En effet, le capteur doit être utilisé pour une mesure prolongée dans le temps pour permettre le suivi continu d'échantillons et ne doit donc pas perturber ces échantillons.En ce qui concerne le procédé de fabrication, il a pour objectif d'être robuste et reproductible pour pouvoir produire des capteurs intégrables dans des chaines de mesure générant d'importants volumes de données normalisées. Le second objectif du procédé de fabrication est la réalisation de capteurs pouvant être réalisés en grandes séries et à bas coût unitaire.Le manuscrit de thèse présente les recherches et déductions menant à la définition de l'architecture du capteur, du procédé de fabrication et des premiers résultats de réalisation. Après trois années de recherche, les travaux de réalisation présentent une structure à base de transistors, utilisant des nanotubes de carbone en tant que canal sensible à l’effet de champ et prédisposé à l’ajout de sélectivité par fonctionnalisation.
