Valorization of marine gastropods in ecotoxicology by studying their genetic diversity and their responses to environmental disturbances

The study of the impact of anthropogenic pollution on living organisms has become a major social issue. In this context, the objective of this thesis was to evaluate the use of gastropods such as limpets (Patellogastropoda) and periwinkles (Caenogastropoda) as indicator organisms of the quality status of their marine environment. Three species were included in the present study: two species of limpets (Patella caerulea, Mediterranean limpet; P. vulgata, Atlantic limpet) and one species of periwinkle (Littorina littorea, common periwinkle). The genetic variability of these species has been assessed by investigating genetic markers (mtDNA: COI, D-Loop or rDNA: ITS) in order to understand how the interconnection networks between different populations are carried out and to evaluate whether distinct genetic variability patterns may exist between different populations. This step made it possible to highlight the genetic mixture and homogeneity of these species along their native distribution areas. A slight genetic differentiation from North to South in the Atlantic is observable with L. littorea and P. vulgata while P. careulea didn’t show any genetic structuring in the Mediterranean (this could be explained in particular by the role of marine currents in the dispersal and passive transportation during the larva stages). An in situ ecotoxicological approach on the Mediterranean limpet (P. caerulea) sampled along the Tunisian coasts [from North to South : Cap Bon (CB), Chebba (C), Kerkennah (K) and Gabès (G)] was carried out by assessing the accumulation of metals (Cd, Cu, Fe, Ni, Pb and Zn) in the water and in the tissues of limpets and by evaluating the antioxidant enzymatic responses (CAT, SOD, GPx) and lipid peroxidation (MDA). L’étude de l’impact des pollutions anthropiques sur les organismes vivants est devenue un enjeu social majeur. Dans ce contexte, l’objectif de ce travail de thèse a été d’évaluer l’utilisation des gastéropodes de type patelles (Patellogastropoda) et littorines (Caenogastropoda) comme organismes indicateurs de la qualité de leur environnement marin. Trois espèces ont été incluses dans ce travail : deux espèces de patelles (Patella caerulea, patelle méditerranéenne ; P. vulgata, patelle d’Atlantique) et une espèce de littorine (Littorina littorea, bigorneau commun). La variabilité génétique de ces espèces a été appréhendée par l’usage des marqueurs génétiques (ADNmt : COI, D-Loop ou ADNr : ITS) afin de comprendre comment s’effectuent les réseaux d’interconnexion entre différentes populations. Cette étape a permis de mettre en évidence l’homogénéité génétique de ces espèces le long de leurs aires de répartition. Une légère différenciation génétique du Nord au Sud en Atlantique est observable pour L. littorea et P. vulgata tandis que P. careulea est sans structuration génétique en Méditerranée (ceci pourrait être notamment expliqué par les courants marins assurant une large dispersion des stades larvaires). Une approche écotoxicologique in situ sur la patelle méditerranéenne (P. caerulea) prélevée le long des côtes tunisiennes [Nord au Sud : Cap Bon (CB), Chebba (C), Kerkennah (K) et Gabès (G)] a été réalisée en déterminant l’accumulation de métaux (Cd, Cu, Fe, Ni, Pb et Zn) dans l'eau et dans les tissus des patelles, en évaluant les réponses enzymatiques biochimiques antioxydantes (CAT, SOD, GPx) et la péroxydation lipidique (MDA).
